SBP-7455 is a potent, selective and orally active ULK1/2 inhibitor with IC50 of 13 nM (ULK1) and 476 nM (ULK2) in ADP-Glo assays, selectively blocks autophagic flux in TNBC cell lines.
SBP-7455 blocks phosphorylation of downstream autophagy target proteins Beclin1 and VPS34.
SBP-7455 is an efficacious tool that can be used to modulate autophagy in vivo.
SBP-7455 is cytotoxic as single agents against TNBC cells with IC50 of 0.3 uM for MDA-MB-468 cells, which is more potent than SBI-0206965.
SBP-7455 promotes apoptosis in TNBC cells under nutrient-deprived conditions.
SBP-7455 reduces autophagic flux and exhibits synergistic cytotoxicity with the PARP inhibitor olaparib against MDA-MB-468 cells, reverses olaparib-induced autophagic flux.
