TM-5441 is a novel potent, orally active PAI-1 (plasminogen activator inhibitor-1) inhibitor that attenuates L-NAME-induced hypertension and vascular senescence in mice.
TM-5441 does not inhibit other SERPINs such as antithrombin III and α2-antiplasmin.
TM-5441 decreases cell viability in several human cancer cell lines with IC50 of 3.58-60.3 uM and induces intrinsic apoptosis at 50 uM.
TM-5441 disrupts tumor vasculature in HT1080 and HCT116 xenotransplanted mice (20 mg/kg daily).
TM-5441 also protects against high-fat diet-induced obesity and adipocyte injury in mice.
