USP30 inhibitor Compound 39 (CMPD-39, USP30inh) is a highly selective inhibitor of USP30 with IC50 of 20 nM in an in vitro assay of enzyme activity, >100-fold selectivity over other DUBs.
CMPD-39 promotes depolarization-dependent ubiquitylation of previously described USP30 substrates (TOMM20, SYNJ2BP).
CMPD-39 enhanced basal mitophagy in treated SHSY5Y-mitoQC cells, enhanced basal pexophagy in treated U2OS-Keima-SKL cells.
CMPD-39 recapitulated the previously described effect for USP30 depletion in cell assays.
