TLR2 inhibitor C-29 is a small molecule TLR2 inhibitor that blocks hTLR2/1 and hTLR2/6 signaling with IC50 of 19.7 uM and 37.6 uM, respectively, in NF-κB reporter assay in HEK293T cells.
TLR2 inhibitor C-29 targets the BB loop pocket of TLR2 TIR domain.
TLR2 inhibitor C-29 inhibits TLR2/1 and TLR2/6 signaling induced by synthetic and bacterial TLR2 agonists in human HEK-TLR2 and THP-1 cells, but only TLR2/1 signaling in murine macrophages, does not inhibit signaling induced by other TLR agonists and TNF-α.
TLR2 inhibitor C-29 blocks robust MAPK activation, reduces NF-κB activation, and prevents P3C-induced degradation of IκBα.
